沙门氏菌(Salmonella)是一种常见的食源性致病菌,对公众健康构成严重威胁。噬菌体是细菌性疾病防控的一种绿色生物制剂。为了筛选广谱高效的噬菌体,以鼠伤寒沙门氏菌D6为宿主菌,从猪粪便样品中分离出1株短尾噬菌体TA2,其基因组不携带耐药和毒力相关基因。TA2可裂解7种不同血清型的沙门氏菌,并且能够抑制细菌生物膜的形成。另外,TA2对温度(≤60℃)、pH(3~12)、10%氯化钠、80%氯仿、乙醇(≤50%)、异丙醇(≤40%)和紫外照射等因素具有耐受性。利用猪肉沙门氏菌污染模型分析发现,TA2能够显著消减猪肉表面沙门氏菌的载量,为噬菌体在食品行业的应用提供了重要数据。
Abstract:Salmonellais one of the common foodborne pathogens,which can cause serious threat to public health.Bacteriophage is a green biological agent against bacterial diseases.In order to screen the phage with broad host spectrum and high effect of lysis,in this study,phage TA2 with extremely short tail was isolated from pig fecal samples using S.typhimurium D6 as the host bacteria.TA2 genome does not carry antibiotic resistance or virulence-associated genes.TA2 can lyse 7different serotypes of Salmonellaand can inhibit the formation of bacterial biofilms.Besides,TA2 exhibits tolerance to temperatures(≤60℃),pH(3-12),10%sodium chloride,80%chloroform,ethanol(≤50%),isopropanol(≤40%)and ultraviolet irradiation.Using the pork model contaminated with S.typhimurium,it was found that TA2 can significantly reduce the Salmonellaload on the surface of pork.This study provides important data for the application of bacteriophages in the food industry.
