兔源奇异变形杆菌耐药性及耐药基因的分析

摘要:

探究兔源奇异变形杆菌的耐药性与耐药基因携带情况。通过聚合酶链式反应(PCR)鉴定分离菌株,运用微量稀释法(Minimum inhibitory concentration, MIC)观察耐药表型,并检测氨基糖苷类、四环素类和磺胺类耐药基因。结果显示,分离菌株HN/Q1和HN/Q2呈现多重耐药,对受试的氨基糖苷类药物(庆大霉素、卡那霉素和阿米卡星)、四环素类药物(四环素和多西环素)、磺胺类药物(复方新诺明、磺胺甲噁唑和磺胺氯吡嗪钠)全部耐药。2株分离菌株均携带aac(6′)-Ib-cr、tetB、suⅠ和suⅡ等耐药基因,HN/Q2还携带aadA2基因。耐药表型与基因型存在一定相关性,磺胺类药物两者符合率达100%,氨基糖苷类和四环素类符合率较低,暗示可能存在其他耐药机制。两株兔源奇异变形杆菌耐药严重,为兔源奇异变形杆菌耐药机制的研究和科学防控提供参考。

Abstract:

This study investigated antimicrobial resistance profiles and associated genetic determinants in rabbit-derived Proteus mirabilis isolates.Through polymerase chain reaction(PCR)-based strain identification and minimum inhibitory concentration(MIC) determination, and the resistance genes of aminoglycosides, tetracyclines and sulfonamides were detected.The results showed that the isolated strains HN/Q1 and HN/Q2 showed multi-drug resistance to the tested aminoglycoside drugs(gentamicin, kanamycin and amikacin);tetracycline drugs(tetracycline and doxycycline);sulfonamides(trimethoprim-sulfamethoxazole, sulfamethoxazole and sulfachloropyrazine sodium) were all resistant to 8 kinds of antibiotics.In addition, the two isolates carried resistance genes such as aac(6′)-Ib-cr,tetB,suⅠ and suⅡ,and HN/Q2 also carried aadA2 gene.There was a certain correlation between drug resistance phenotype and genotype.The coincidence rate of sulfonamides was 100%,and the coincidence rate of aminoglycosides and tetracyclines was low, suggesting that other drug resistance mechanisms may exist.These results demonstrate that the two strains of Proteus mirabilis from rabbits in this study have serious drug resistance.This study provides theoretical support for further research on the drug resistance mechanism of rabbit-derived Proteus mirabilis,scientific prevention and control, and rational drug treatment.
